Sequence-Dependent Viscoelasticity of Biomolecular Condensates

Biomolecular condensates are complex liquid-like subcellular organelles that segregate client molecules from their environment for intracellular storage, signaling, and the amplification of biochemical processes. The material properties a biomolecular condensate essential to its physiological function. Emerging reports suggest on liquid-end viscoelastic spectrum functional, whereas solid-end often linked pathological states. However, molecular driving forces underlying origin viscoelasticity is currently unknown, except qualitative assessment asymptotic cases where purely liquid or solid in nature. Here, using optical tweezers, we implement active passive microrheology quantitatively assess series artificial peptide-RNA condensates. Our experiments fluids with time-dependent viscous elastic moduli, extent elasticity these precisely encoded by amino acid sequence composition patterning polypeptide chains.
